Atebimetinib's Phase 3 MAPKeeper 301 trial tests whether striking Phase 2a OS data in first-line PDAC translates to a registrational win in mid-2028

The most likely failure modes are a Phase 2-to-Phase 3 translation gap: the Phase 2a cohort was small (N=55 at the final readout) and single-arm, making it susceptible to selection bias and regression to the mean in a randomized setting. Acquired resistance mechanisms through RAS pathway reactivation — already tracked via ctDNA in the Phase 2a — could blunt the durable OS benefit, and the addition of a formal control arm may reveal that patient selection or site effects inflated the Phase 2a OS numbers.
